ABSTRACT

ObjectiveTo observe the regulation of Qigongwan on the expression of proliferation and apoptosis-related factors programmed cell death 4 (PDCD4) and proliferating cell nuclear antigen (PCNA) in ovarian granulosa cells (GCs) in patients with polycystie ovarian syndrome (PCOS) infertility with phlegm-dampness syndrome, and to explore the effect of Qigongwan on the quality of oocytes and embryonic development potential. MethodSixty-six patients with PCOS with phlegm-dampness syndrome who underwent in vitro fertilization and embryo transfer (IVF-ET) were randomly selected and divided into an observation group (Qigongwan + western medicine) and a control group (western medicine), with 33 patients in each group. Antagonist regimen was used to promote ovulation in the two groups. The observation group was given Qigongwan one cycle before IVF based on the treatment of conventional western medicine, while the control group was not given Chinese medicine. The improvement of phlegm and dampness syndrome, the dosage and the number of days of using gonadotropins (Gn), the levels of luteinizing hormone (LH), estradiol (E2), and progesterone (P) on the day of human chorionic gonadotropin(HCG) injection, the 2PN fertilization rate, and the high-quality embryo rate of patients in the two groups were compared. Real-time polymerase chain reaction (Real-time PCR) and Western blot technology were used to detect the expression of PCNA and PDCD4 in GCs. ResultAs compared with groups before treatment, the score of phlegm-dampness syndrome in both groups was significantly lower (P<0.01). The score of phlegm and dampness syndrome in the observation group was significantly lower than that of the control group (P<0.01). As compared with the control group, the levels of LH, E2, and P in the observation group was higher, but only the difference in the level of E2 was statistically significant (P<0.01). The 2PN fertilization rate [82.25% (556/676) vs 69.92% (365/522), χ2=25.172, P<0.01] and high-quality embryo rate [44.19% (190/430) vs 34.23% (102/298), χ2=7.266, P<0.01] in the observation group were significantly higher than that of the control group (P<0.01). As compared with the control group, the mRNA and protein expression of PDCD4 in ovarian GCs was down-regulated in the observation group and that of PCNA was up-regulated (P<0.05). ConclusionBy down-regulating the expression of PDCD4 and up-regulating the expression of PCNA, Qigongwan may interfere with follicle development, adjust hormone levels, improve the symptomatic manifestations of patients with PCOS with phlegm-dampness syndrome, inhibit the apoptosis of GCs, and promote growth, thus improving the quality of oocytes and embryonic development potential.

ABSTRACT

The fundus lesions caused by high myopia (HM) often lead to irreversible visual impairment or even blindness. However, the pathogenesis of HM and its fundus lesions is still unclear, the intraocular fluid detection technology of micro samples has brought new prospects for the early diagnosis, monitoring and intervention of the fundus lesions. The molecules associated with HM are various and functionally diverse, intermolecular interactions are staggered and the specific mechanism is complex. With the development of intraocular fluid detection technology, while gradually revealing the role of each molecule in the pathogenesis of HM, it is expected to successfully assist clinical work in the future, providing outpost markers for the progress of myopia and targets for early intervention, or providing a new therapy choice for HM fundus lesions at the molecular level targeting pathogenesis, which is expected to provide more accurate and effective treatment for HM patients in the future.

ABSTRACT

Since epileptic seizure is unpredictable and paroxysmal, an automatic system for seizure detecting could be of great significance and assistance to patients and medical staff. In this paper, a novel method is proposed for multichannel patient-specific seizure detection applying the earth mover's distance (EMD) in scalp EEG. Firstly, the wavelet decomposition is executed to the original EEGs with five scales, the scale 3, 4 and 5 are selected and transformed into histograms and afterwards the distances between histograms in pairs are computed applying the earth mover's distance as effective features. Then, the EMD features are sent to the classifier based on the Bayesian linear discriminant analysis (BLDA) for classification, and an efficient postprocessing procedure is applied to improve the detection system precision, finally. To evaluate the performance of the proposed method, the CHB-MIT scalp EEG database with 958 h EEG recordings from 23 epileptic patients is used and a relatively satisfactory detection rate is achieved with the average sensitivity of 95.65% and false detection rate of 0.68/h. The good performance of this algorithm indicates the potential application for seizure monitoring in clinical practice.

ABSTRACT

Objective To observe late effect of 60Co radiation accident on eye lens in the victims.Methods Medical observations of eye lens were performed on ten victims accidentally exposed to 60Co sources in four radiation accidents that have occurred from 1986 to 2000 in Henan Province.Pathological changes of the eye lenses were examined by using slit lamp microscopy after mydriasis with compound tropicamide.Results Of these ten victims, Liang in a radiation accident in Kaifeng (in 1986), Mei in Xinxiang (in 1999) and Xu in Xuchang (in 2000) all had typical radiation-induced cataract 2, 3 and 6 years after irradiation, respectively.Follow-up observation of the lens showed the punctate and/or granular opacities present in the eye posterior subcapsular of Yan in Kaifeng accident, Jie in Zhengzhou (1987), and Tian, Yong and Yi in Xinxiang accident, featuring the early changes of radiation-induced cataract, but the posterior subcapsular opacities were not observed in Wang and Min in Xinxiang accident.Conclusions Focus should be on the eye lens as the target organ of radiation exposure in long-term follow-up of victims accidentally exposed to radiation source.Severity of the lens opacity induced by ionizing radiation is closely associated with radiation doses.

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the effect of combined immunization with recombinant Sap2 and dendritic cells (DCs) against systemic Canddida albicans infection.</p><p><b>METHODS</b>We constructed a prokaryotic expression vector carrying Sap2 of Candida albicans to obtain Sap2 protein. Murine DCs were sensitized by pulsing with Candida albicans spores and rope. Five groups of mice were immunized with recombinant Sap2 protein and sensitized DCs, sensitized DCs, naive DCs, Sap2, or PBS alone for 3 times, and the effect of the immunization against systemic Candida albicans infection were assessed by observing the survival of the mice, detecting the percentages of CD4⁺ and CD8⁺ cells, CFU in the kidney homogenate, and examining renal pathologies.</p><p><b>RESULTS</b>Immunization with Sap2 and sensitized DCs and with DCs or Sap2 alone all prolonged the mouse survival and produced obvious effect in renal protection and immune enhancement, but such effects were more obvious with the combined immunization.</p><p><b>CONCLUSION</b>Combined immunization with Sap2 protein and DCs offers strong immunoprotection against systemic Candida albicans infection in mice, which provides experimental evidence for the development of new combined vaccines for immunoprotection.</p>

ABSTRACT

Objective To explore whether low-dose occupational ionizing radiation exposure can affect the prevalence of lens opacity.Methods Annual occupational health checkup data of 1 007 radiation workers was taken from a provincial medical institution for the purpose of statistical analysis.Logistic regression analysis was used to estimate occupational exposure odds ratios (OR) of lens opacity,adjusted for age,gender and length of service.Eye lens opacity was grouped into cortical,nuclear and posterior subcapsular opacity according to the position of the opacity occurrence site.Opacity occurred in any one of the both eye lens was recorded as turbidity.Results Only 730 cases coupled with complete information could be used in the statistical analysis.The rate of lens opacity for all the radiation workers was 10.27%.The rates of lens opacity by exposure group were estimated to be 9.07% for radiation diagnosis and therapy group,11.11% for intervention group,18.18% for nuclear medicine group,and 9.33% for industrial application group,respectively.Compared with those in the radiation diagnosis and therapy group,the workers engaged in intervention medicine,or nuclear medicine,were more likely to suffer from the lens opacity in posterior subcapsular position.The OR and its 95% confidence intervals were 3.00 (1.23-7.33),4.12 (1.68-10.11) for the workers in intervention medicine or nuclear medicine group.Conclusions Medical radiation workers,who were exposed to long-term low-dose of ionizing radiation,especially those who engaged in intervention or nuclear medicine,were at significantly higher risk to develop lens opacity.

ABSTRACT

Objective To compare the treatment effects of senile intertrochanteric fracture with the dynamic hip screw(DHS),proximal femur locking compression plate (LCP),anti-spin femur proximal femoral nail(PFNA).Methods Ninety cases of femoral rotor fracture patients were treat in our hospital from September 2010 to March 2012 by DHS(n =30),LCP(n =30) and PFNA(n =30) treatment of femoral intertrochanteric fractures.We compared the surgical time,blood loss,fracture healing time,postoperative complications and hip function scores.Results In DHS,LCP and PFNA group,the blood loss ((287.1 ± 72.0) ml,(214.9 ± 65.8)ml,(85.0 ± 33.6) ml,F =88.7,P < 0.05),the operative time ((1.51 ± 0.07) h,(1.49 ± 0.04) h,(0.70 ±0.10) h,F =445.8,P < 0.05),and both were significant difference.But the fracture healing time ((15.2 ±1.7) weeks,(17.0 ± 1.4) weeks,(15.9 ± 1.8) weeks,F =1.4,P > 0.05),there was no significant difference.The PFNA group had a good hip Sander' score of 96.7%,better than other two groups (87.5%、96.8%) (x2 =39.06,P <0.05).The weight-beating time of PFNA group is earlier than the other two groups ((1.00±0.36) cm,(0.80±0.12) cm,(0.60±0.19) cm,F=30.4,P<0.05).Conclusion PFNAinthe treatment of femoral intertrochanteric fracture has advantages in minimally invasive,less blood loss,and help patients go through the perioperative period.

ABSTRACT

AIM: To establish the method for quality control of Yijingbushen Granules(Herba Epimedii, Semen Cuscutae, Radix Paeoniae Alba, Rhizoma Curcumae, etc.). METHODS:Semen Cuscutae, Radix Paeoniae Alba and Rhizoma Curcumae in this medicine were identified by TLC; The content of icarrin in this medicine was determined by HPLC;The acetonitrile-2%acetum(30∶70) solution was used as the mobile phase. The flow rate was 1.0mL?min -1. The detection wavelength was set at 270nm. RESULTS:Semen Cuscutae, Radix Paeoniae Alba and Rhizoma Curcumae, could be identified by TLC on lichrospherC 18 column. For Icarrin, the linear range was from 4.32 to 21.60?g, and the average recovery was 98.69% with RSD of 1.13%. CONCLUSION:The method is simple, reliable, accurate and can be applied to the quality control of the preparation.
